Semaglutide (GLP-1) research overview

Semaglutide is a modified, acylated analogue of human glucagon-like peptide-1 (GLP-1). This page summarizes primary research on its molecular design, receptor engagement, and selected preclinical models without turning those findings into human-use guidance.

What is semaglutide?

The 2015 discovery paper described semaglutide as a GLP-1 analogue with two amino-acid substitutions and a fatty-diacid modification at lysine 26. The design was evaluated for GLP-1 receptor activity, albumin binding, metabolic stability, and prolonged exposure in experimental systems.

How has GLP-1 receptor engagement been studied?

The discovery program measured receptor potency alongside albumin affinity and stability. A 2021 cryo-electron microscopy study visualized semaglutide bound to the GLP-1R–Gs complex and reported peptide interactions similar to native GLP-1 together with differences in motion. Structural observations explain binding geometry, not clinical outcomes.

What do selected preclinical studies add?

A 2020 study used mice and rats to investigate neural access, GLP-1 receptor distribution, food intake, and energy expenditure. Those findings support mechanistic hypotheses in the tested animal models; they are not dosing guidance or proof that the same effects occur in people.
