MOTS-c research overview
MOTS-c is a 16-amino-acid mitochondrial-derived peptide first reported in 2015. This page summarizes what peer-reviewed laboratory studies report, the models they used, and what those findings do not establish.

What is MOTS-c?
MOTS-c stands for mitochondrial open reading frame of the 12S rRNA-c. The original 2015 report described a short open reading frame within mitochondrial 12S rRNA that encodes a 16-amino-acid peptide. That work examined endogenous MOTS-c and experimental peptide exposure in cultured cells and mouse models.
Mechanisms investigated in laboratory studies
The 2015 experiments reported changes involving folate and purine pathways together with AMPK-associated signaling. A 2018 cell-culture study reported stress-associated movement of MOTS-c into the nucleus and changes in stress-responsive gene expression. These mechanistic findings are not proof of clinical benefit.
MOTS-c and exercise research
A 2021 paper combined mouse experiments, cell studies, and measurements from ten young male participants. The human portion observed endogenous MOTS-c around an exercise session; it did not test a marketed MOTS-c product as a treatment. “Exercise mimetic” should not be read as evidence that a research product replaces exercise in people.
